Covalent Inhibitory Probes of Human Cysteine Cathepsin L and B

Cysteine cathepsins are a small group of eleven human enzymes that are critically important in maintaining homeostasis, growth, differentiation, and survival. One of the main challenges of cathepsin biology is in assigning specific function to individual enzymes in specific cellular context. This is primarily because of functional redundancies and overlapping substrate specificities. Since newly synthesized cellular cathepsins are subject to post-translational modifications, thereby leading to alteration of their activities (and hence function), investigative tools are needed for activity-based interrogation of cellular function in live cells. This work focuses on development of selective clickable and tagless activity-based probes (catABPs) of two closely related cysteine cathepsins, cathepsin L and B. The developed catABPs will be utilized in differential and quantitative mapping of cathepsin B and L activity profile in live cells from distinct tissue origins.
