A photochemical rearrangement to construct palau’amine
Defining 'complexity' when referring to molecules is, well… complex. Complexity certainly increases with the number of atoms and chiral centers, but the stability of the molecule, its internal strain, or its sensitivity to air, moisture, light or other stimuli also contribute. To organic chemists, complexity is often related to the difficulty in preparing such structures from known transformations. The photochemical rearrangement of N-mesyloxylactams is used to convert a 5-6 trans-fused ring system into its highly strained 5-5 trans-fused homologue in high yield. A short background to the photochemical rearrangement will be given and this key reaction will be presented in the context of a synthesis of a core fragment of palau’amine. Our efforts to make an analogous thermal rearrangement directly from non activated amide will also be discussed.
